BT549 human breast cancer spheroids were embedded in TissueSpec® Lung dECM Hydrogel to model cell invasion in a secondary site of metastasis and for subsequent transcriptomic analysis. Breast cancer spheroids were imaged at Day 0, 3, and 7. Yellow arrowheads denote tumor cells that have disseminated from the spheroid to initiate invasion into the surrounding lung dECM microenvironment. Scale bar = 100 µm.
TissueSpec® Lung dECM Hydrogel Kit: RNA-seq analysis on BT549 human breast cancer spheroids embedded in TissueSpec® Lung dECM Hydrogel or Matrigel
BT549 human breast cancer spheroids were embedded in either TissueSpec® Lung dECM Hydrogel or Matrigel for 3 days to assess the influence of two distinct extracellular matrix environments on tumor cell gene expression. After incubation, RNA was extracted from BT549 spheroids for transcriptomic analysis and a total of 3,181 genes were found to be differentially expressed (p ≤ 0.05) between the two conditions. Of these, 453 genes were significantly upregulated and 1,259 genes were significantly downregulated (p ≤ 0.01) in the TissueSpec® Lung dECM Hydrogel compared to Matrigel.
TissueSpec® Lung dECM Hydrogel Kit: Differential gene expression analysis on BT549 human breast cancer spheroids embedded in TissueSpec® Lung dECM Hydrogel or Matrigel

Primary normal human bronchial epithelial cells cultured on thin layers of TissueSpec® Lung dECM Hydrogel (6mg/mL) or Matrigel for 10 days. Normal lung epithelial markers p63 (red) and ZO-1 (green) were used.
